Histological Studies on Mast Cells of the Urinary System of Cattle, Sheep and Goats
2 Ankara Üniversitesi, Veteriner Fakültesi, Histoloji-Embriyoloji Anabilim Dalı, Ankara - This study was conducted with the aim of revealing of the morphology and numerical distribution of mast cells of the urinary system of cow, sheep and goats at light microscopic level and investigating the heterogeneity of mast cells by using different fixation solutions. In the study, tissue samples which were taken from the urinary system of 10 healthy cow, sheep and goats were used as the study material. Tissue samples were fixed with both 10% formaldehyde and isotonic formaldehyde acetic acid (IFAA) solutions. Toluidine blue and alcian blue /safranin O (AB/SO) combined staining methods were applied for the mast cells. As a result of the examination on the light microscopic level, it was observed that mast cells have spherical, ovoid and elongated, depending on their location, shapes and different sizes. It was founded that IFAA fixed tissues were contained more mast cells than those tissues fixed with the 10% formaldehyde. Tissue samples which were taken from the urinary system of cattle, sheep and goats stained with the AB/SO combined staining method and AB(+)/SO(-) mast cells have been found in the samples. In the serial sections stained with Toluidine blue to determine the distribution of mast cells arithmetic mean was used. The difference in average number of mast cells per mm2 between the IFAA and the 10% formaldehyde fixed tissues was statistically significant (P<0.01).
