Determination of Polymorphism in Acyl-coenzyme A Dehydrongenase Gene (g.2885C>A) by ARMS-PCR Methods in Some Cattle Breeds Raised in Turkey

1 Akdeniz Üniversitesi Ziraat Fakültesi Zootekni Bölümü, Antalya - Acetyl coenzyme A dehydrogenase (ACADVL) is a protein that plays an important role in oxidation of long chain fatty and releasing energy. SNP (g.2885C>A; Pro236Thr) on the ACADVL dehydrogenase gene located on chromosome 19 in cattle is associated with some growth traits (chest width, chest depth and hip width). Individuals with AA genotype have superior growth traits than individuals with AC and CC genotypes. In this study was aimed to determine polymorphism on ACADVL gene (g.2885C>A) in Holstein (SA), Anatolian Black (YK) Turkish Grey Steppe (BI), and East Anatolian Red (DAK) cattle breeds raised in Turkey by ARMS-PCR method. In this study was used totally 210 samples obtained from SA (64 samples), YK (54 samples), BI (48 samples) and DAK (44 samples) cattle breeds. As a result of ARMRS-PCR analyzes for ACADVL (g.2885C>A) gene SA breed was found to be monomorphic (CC-307- 152 bp) while BI, DAK and YK cattle breeds were found to be polymorphic (AC: 307-211-152 bp and CC: 307-152 bp). Frequency of AC genotype were calculated as 0.063, 0.045 and 0.093 while frequency of CC genotype were calculated as 0.937, 0.955 and 0.907 in BI, DAK and YK cattle breeds, respectively. The polymorphism was shown for the frst time on ACADVL (2885C>A) gene in SA, YK, BI and DAK cattle breeds. AA genotype could not be detected in studied cattle breeds.
